确实因为where 和 what 是疑问副词和疑问代词的区别在这两句句尾才一个没有介词一个有。 Be made of 是古定搭配，of不能省。
但也有的情况下，疑问副词前用介词：
Where do you come from?
你是哪儿人?
介词一般放在名词之前。但它后面的介词宾语是疑问代词,疑问副词或者关系代词时，这些词提到了前面而只剩下介词在后了。
where is this pen made? 原句：This pen is made in China.（对in China提问就成了上句,对地点提问，in也划线）
what is this pen made of? 原句：This pen is made of plastic.(对plastic提问就成了上句，对物提问，of没划线）
