ABC of DNA and its application

The ABC of DNA

Release date: November 03, 1998

After a nationwide search for Britain's most innovative products, a DNA test kit for schools developed by The University of Reading's National Centre for Biotechnology Education has been granted Millennium Product status by the Design Council. The news was announced by Trade and Industry Secretary Peter Mandelson at the CBI Conference in Birmingham on Monday 2 November. The centre's invention is already used by a quarter of UK secondary schools and colleges, and is sold throughout the world. It has now been short-listed for inclusion in the Millennium Dome in Greenwich.

John Schollar, who with Dean Madden was responsible for the invention said "Our inexpensive DNA kit allows pupils to produce the familiar bar-code like patterns that are seen on genetic fingerprints. It has been designed with ease-of-use and safety in mind, yet gives results comparable to those produced by conventional equipment costing many thousands of pounds." "Develoments in modern genetics affect everyone" said Dean Madden. "It is vital that school students learn about modern DNA technology and its potential implications, so that society can benefit from it while ensuring that its use is properly controlled. We hope that our equipment will help teachers to bring this exciting area of technology to life." Local injection-moulding company, Firma Nicand in Woodely, custom-made one of the key components in the kit.

exterion Slide H produces exceptionally good spot morphologies and consistent spot sizes for oligonucleotide microarray applications. The hydrogel coating of Nexterion Slide H das an excellent loading capacity, which result in a better signal-to-background ratio when compared to the leading 3D slide and conventional coatings (such as silanized slides). This was demonstrated with dual-color hybridization experiments following the recommended protocols and using amine-modified oligonucleotides (50mers) as probes.

Signal Intensity

Nexterion Slide H provides excellent and reproducible signal-to-background (S/B) ratios for oligonucleotide microarray applications when compared to the leading 3D slide. Nexterion Slide H performs particularly well with short amine-modified oligonucleotides (=< 25mers), since the special coating allows highly specific end-point attachement of oligonuclaotide probes (see section Specificity of attachment) The figure below shows normalized S/B ratios for an oligonucleotide-oligonucleotide assay, with oligonucleotides being immobilized in various spotting solutions and hybridized with fluorescence-labeled complementary oligonucleotides (probe and target 16mers). Slides were processed according to the protocol recommended by the manufacturer of the leading 3D slide.
Specificity of attachement

The functional groups within the multicomponent 3D coating matrix of Nexterion Slide H are highly specifc for amine-modified oligonucleotides. This is especially advantageous when short oligonucleotides are immobilized (e.g. for mutation analysis or SNP detection assays). The figure shows normalized S/B ratios for an oligonucleotide-oligonucleotide assay, with probes (with and without amine-modification) being immobilized in various spotting solutions and hybridized with fluorescence-labeled complementary oligonucleotides (probe and target 16mers). Slides were processed according to the protocol recommended by the manufacturer of the leading 3D slide.
